Raj,
The #:# notation, like "eth0:0", implies an alias (secondary IP).

The #.# notation, like "eth0.0", implies a vlan (use vconfig).

On RHEL/CentOS like systems, you can add IPADDR2= and PREFIX2= (or
NETMASK2=) lines to the config file ifcfg-eth0 to add the IP at boot
(assuming NetworkManager isn't mangling anything).
Have you tried, that so easy to test

ifconfig eth0:1 172.16.1.10 netmask 255.255.0.0

Then try to another ip on that network. If it doesn't work then

ifconfig eth0:1 down

Now I am thinking it might not work because of the vlans.
Is it possible to have a NIC with multiple addresses on different VLANs?
Example:

eth0.0 == 10.1.1.10
eth0.1 == 172.16.1.10
