You've probably locked yourself out in some way. I was just trying to play around with recreating your problem and locked myself out by adding hosts deny = all to my smb.conf file. I know this doesn't recreate your problem but it does show that it's easy to screw yourself up without meaning to.
Try looking at your /etc/smb.conf file for obvious errors. If that doesn't work, email on of us your smb.conf file for review. I would like to know what is causing your problem in case it happens to me in the future. BTW I'm one file server and one PDC away from obliterating MS from my server room altogether, so this is more than a little interesting to me. :)
